For a positive control, 2 × 105 NK92 cells (overnight rested without IL-2) were pre-incubated for 1 h with 200 ng/ml C1.7 anti-2B4 antibody and subsequently incubated with untransfected K562 target cells for 16 h. Assays were conducted in triplicates with all proper standards and controls. Inhibitor treatment of cells.  NK92 cells were rested overnight without IL-2 and then pre-incubated with functional concentrations of various pharmacological inhibitors for an appropriate period of time prior to initiation of IFN-γ release assay. Inhibitors and concentrations employed were 20 μg/ml Actinomycin D, 10–50 μm SB203580, 50–100 μm PD98059, 1 nm ascomycin, 10 μm PP2, 25 μm LY294002, and 1 μm Bisindoylmaleimide I. Inhibitors were dissolved in DMSO. NK92 cells without the inhibitors were incubated with an equal amount of DMSO that served as a control.
